My prediction is when Blair asks him to kiss that guy, Chuck will either say one of two things to Blair:
1) "It's not like I haven't done it before..." (or some variation of that, which would indicate that he is bi), or
2) "Only if you agree to kiss Serena".
What else could he possibly say??

I don't think he saying to Blair that he can't do that relationship anymore. The writers already said that Chair will not be breaking up this season. I think he is saying it to Blair because maybe Blair wanted Chuck to lead on the guy so Blair could get something good out of it for NYU? Oh well. The writers always make us think that it something terrible when it reality it really isn't.

Now I think he will probably make a stupid comment, something funny and irrelevant. Maybe he's not bi at all.
Or maybe, maaaybe, he asked the guy to help Vanessa instead of helping Blair, and then he says something mean to Blair and call Vanessa to tell her she got it. That would be nice, I mean, Blair would have to get over herself and be less stupid and manipulative from then on.
